Abstract

Stiren je monomer koji se koristi u proizvodnji CD kutija, kućanskih aparata, guma, dijelova za brod i mnogih drugih proizvoda, a zbog široke upotrebe u industriji može dospijeti u okoliš. Zbog štetnosti stirena i stiren-oksida velika se važnost pridaje mikroorganizmima koji su ih u mogućnosti ukloniti. Korištenje živih bića za uklanjanje otpada iz okoliša naziva se bioremedijacija. Jedan od organizama koji je u mogućnosti metabolizirati stiren je bakterija Pseudomonas putida. Radi se o bakteriji koju u prirodi nalazimo u tlu. Osim za bioremedijaciju veliki interes za organizme poput P. putide postoji i zbog njihove moguće primjene u industriji. Biotransformacijom stirena u stiren oksid uz korištenje enzima stiren monooksigenaze porijeklom iz Pseudomonas putida dobiva se produkt velike enantiomerske čistoće. Biotransformacija stirena u (S)-stiren oksid provedena je u kotlastom reaktoru pomoću genetski modificirane bakterije Pseudomonas sp. VLB120ΔC koja proizvodi enzim monooksigenazu. Indukcija enzima je provedena dodavanjem stirena nakon završetka rasta bakterije. Rast biomase proveden je u Erlenmayer tikvicama. U radu je ispitan utjecaj promjene uvjeta indukcije na količinu proizvedenog (S)-stiren oksida.

Abstract (english)

Styrene is a monomer commonly used in manufacturing CD casings, home appliances, rubber, ship parts and others, and due to its widespread industrial usage accidental emissions into the environment are quite likely. Due to the potential environmental damage of styrene and styrene-oxide contaminations microorganism that can reverse the damage are very important. Using living organisms to remove waste from the environment is called bioremediation. One of the organisms capable to metabolize styrene is the bacterial strain of Pseudomonas putida. In nature this bacteria can be found in the soil. Aside from bioremediation purposes, there's a great interest towards P. putida and similar organisms because of their potential industrial applications. Using the styrene monooxygenase enzyme ( which is produced by P. putida) to produce styrene oxide yields a product with large enantiomeric excess. Biotransformation of styrene into (S)-styrene oxide was conducted in a batch reactor using the genetically modified bacteria Pseudomonas sp. VLB120ΔC produces the styren monooxygenase enzyme. The enzyme was cultivated by adding styrene after the end of inoculation period. Biomass growth was conducted in Erlenmeyer flasks. This work examined the effects of different cultivation parameters in regards to the yield of (S)-styrene oxide.
