Abstract

Aditivne tehnologije su zbog raznih prednosti i jednostavnosti u kratkom vremenu našle svoju primjenu u nebrojenim industrijama. Između ostalog, omogućeno je 3D tiskanje tableta iz prethodno pripravljenih filamenata. Cilj ovog rada bio je proizvesti filament koji sadrži djelatnu tvar za 3D tiskanje tablete. U eksperimentu su za pripravu filamenta korišteni poli(vinil-alkohol) (PVA), dronedaron-hidroklorid kao djelatna tvar te sorbitol kao plastifikator. Pripravljene su dvije mješavine koje se razlikuju po veličini čestica PVA. Jedna mješavina sadrži PVA veličina čestica manjih od 2500 μm (F2500), a druga PVA veličina čestica manjih od 710 μm (F710). Filamenti su dobiveni procesom ekstruzije pri čemu je temperatura bila ključan parametar. Testiran je utjecaj temperature na proces ekstruzije i zaključeno je da pripravljenim mješavinama najbolje odgovara temperatura od 145 °C. Nakon ekstruzije filamenti su otopljeni u vodi i metanolu, otopine su filtrirane te podvrgnute analizi na UV/Vis spektrofotometru. Prethodno je iz standardnih otopina dobiven umjerni pravac pomoću kojeg je određena koncentracija dronedaron-hidroklorida. Iz koncentracije je dobivena vrijednost udjela dronedaron-hidroklorida u filamentu. Osim udjela dronedaron-hidroklorida u filamentu, određeni su i promjeri filamenata. Rezultati su pokazali značajna odstupanja udjela dronedaron-hidroklorida između pripravljene mješavine i dobivenog filamenta. Udio dronedaron-hidroklorida u obje početne mješavine bio je 9,1 %, a analizom filamenata određeno je odstupanje od 52,3 % za filament F710 i 57,7 % za filament F2500. Odstupanje od ciljanog promjera filamenta (1,75 mm) iznosilo je 16,2 % za filament F710 i 17,8 % za filament F2500. Iz dobivenih odstupanja vidljivo je da je mješavina koja sadrži čestice PVA manje veličine pogodnija za proizvodnju filamenata procesom ekstruzije zbog boljeg miješanja s dronedaron-hidrokloridom.

Abstract (english)

Additive manufacturing found its application in countless industries because of its simplicity and many advantages. It enabled 3D printing of tablets from custom created filaments. Purpose of this paper was to make a filament for 3D printing that contains active pharmaceutical ingredient. PVA (poly(vinyl-alcohol)) was used with dronedarone hydrochloride as active pharmaceutical ingredient and sorbitol as plasticizer. Two mixtures have been prepared that differ by particle size of used PVA. One mixture contains PVA with particle size smaller than 2500 μm (F2500), and the other contains PVA with particle size smaller than 710 μm (F710). Filaments were produced by extrusion process and the key parameter was temperature. Temperature influence has been tested and it has been concluded that the best temperature is 145 °C. After the extrusion process, filaments have been dissolved in water and methanol; solutions have been filtered and analyzed by UV/Vis spectrophotometry. Caliber line has been made using standard solutions, and with it, concentration of dronedarone hydrochloride has been determined. Using the concentration, mass percentage of dronedarone hydrochloride in the filament has been determined. Apart from dronedarone hydrochloride content in the filaments, filaments diameter have been determined. Results show significant deviation of dronedarone hydrochloride content in prepared mixture and produced filament. Dronedarone hydrochloride content in both prepared mixtures was 9.1 wt.%. Deviation of content for filament F710 was 52.3 wt.% and 57.7 wt.% for filament F2500. Filament diameter differs from the targeted value of 1.75 mm. Deviation in diameter is 16.2 % for filament F710 and 17.8 % for filament F2500. Results show that PVA with smaller particle size is better for producing filaments by extrusion process because of better blending with dronedarone hydrochloride.
