Abstract

Oksidoreduktaze su vrsta enzima koji za svoje katalitičko djelovanje zahtijevaju prisutnost koenzima. Zbog visoke cijene koenzima važno je istražiti učinkovit način regeneracije koenzima kako bi se smanjili troškovi u industrijskim procesima. U ovom radu ispitana je mogućnost regeneracije koenzima NADP+ reakcijom redukcije acetona katalizirane ketoreduktazom. Reakcija redukcije acetona je ravnotežna reakcija. Kinetika ketoreduktaze u reakciji redukcije acetona ispitana je metodom početnih brzina za naprednu i povratnu reakciju, redukciju acetona i oksidaciju izopropanola. Kinetika obje reakcije je opisana dvosupstratnom Michaelis-Menteničinom kinetikom. Na temelju postavljenog matematičkog modela i procijenjenih parametara, pomoću programskog paketa Scientist simulirana je vremenska ovisnost koncentracije reaktanata i produkata reakcije redukcije acetona u kotlastom reaktoru. Dobivena ravnotežna konverzija acetona iznosila je 54,24 %.

Abstract (english)

Oxidoreductases belong to a class of enzymes which require coenzyme for catalytic activity. Because of a high cost of coenzyme, it is very important to investigate an efficient method of coenzyme regeneration to reduce these costs in industrial processes. In this work, the possibility of regeneration of coenzyme NADP+ was examined by the reaction of acetone reduction catalyzed by ketoreductase. The reaction of acetone reduction is reversible. Ketoreductase was kinetically characterized by determining the initial enzyme reaction rate for both directions of reversible reaction - acetone reduction and isopropanol oxidation. Kinetics of both reactions is described with a two-substrate Michaelis-Menten kinetics. The time dependent concentration of reactants and products was simulated by Scientist, softvare program, for a batch reactor, based on mathematical model and estimated parameters. The equilibrium conversion of acetone was 54,24 %.
