Abstract

Niskotemperaturna eutektička otapala vrsta su otapala koje karakterizira značajno sniženje tališta u usporedbi s talištima čistih komponenti od kojih su sastavljena. S obzirom na dokazana izvanredna svojstva niskotemperaturnih eutektičkih otapala i njihovu ekološku prihvatljivost, postala su obećavajuća alternativa dosadašnjim otapalima. U farmaceutskoj industriji niskotemperaturna eutektička otapala mogu biti korištena u svrhu poboljšanja topljivosti, permeabilnosti i apsorpcije djelatne tvari. U ovom radu pripremljena su i karakterizirana niskotemperaturna eutektička otapala miješanjem djelatne tvari fosamprenavir kalcija s različitim netoksičnim komponentama u određenom molarnom omjeru. Svrha rada je dobiti terapeutsko niskotemperaturno eutektičko otapalo koje ima isto terapijsko djelovanje kao i djelatna tvar od koje je pripremljeno, ali poboljšana svojstva. Kako bi se dobio uvid u karakteristične fazne i temperaturne promjene u otapalu kao i potvrdio nastanak vodikovih veza među pomiješanim komponentama, provedena je analiza na diferencijalnom pretražnom kalorimetru i infracrvenom spektrofotometru s Fourierovom transformacijom. Ispitana je maksimalna topljivost, tijek oslobađanja djelatne tvari iz terapeutskog niskotemperaturnog eutektičkog otapala i permeabilnost. Kinetičko ponašanje oslobađanja opisano je različitim matematičkim modelima.

Abstract (english)

Deep eutectic solvents are a type of solvents characterized by a significant lowering of the melting point compared to the melting points of the pure components from which they are composed. Given the proven outstanding properties of deep eutectic solvents and their environmental acceptability, they have become a promising alternative to current solvents. In the pharmaceutical industry, deep eutectic solvents can be used to improve the solubility, permeability and absorption of the active pharmaceutical ingredients. In this paper, deep eutectic solvents were prepared and characterized by mixing the active pharmaceutical ingredient fosamprenavir calcium with different non-toxic components in a certain molar ratio. The purpose of the work is to obtain a therapeutic deep eutectic solvent that has the same therapeutic effect as the active pharmaceutical ingredient from which it was prepared, but with improved properties. In order to gain insight into the characteristic phase and temperature changes in the solvent as well as to confirm the formation of hydrogen bonds between the mixed components, an analysis was performed on a differential scanning calorimeter and an infrared spectrophotometer with Fourier transformation. Maximum solubility, the release of the active pharmaceutical ingredient from the therapeutic deep eutectic solvent, and permeability were tested. The kinetic behavior of the release is described by different mathematical models.
