Abstract

Upotreba enzima danas je rasprostranjena u mnogim industrijskim granama, posebice onima koje pripadaju područjima biotehnologije, kemijskog inženjerstva i procesnog inženjerstva. Jedan od enzima koji posljednjih desetljeća ima široku primjenu u industriji je endo-1,4-ksilanaza, koji se u svakodnevnoj praksi često skraćeno naziva ksilanaza. Veliki broj mikroorganizma proizvodi enzim ksilanazu, a najboljim izvorom ovog enzima smatraju se nitaste gljive. Usprkos tome, brojna su istraživanja usmjerena pronalaženju novih izvora ksilanaze. Kad se odgovarajući izvor enzima ksilanaze pronađe, te ovaj enzim identificira i izolira, glavni ograničavajući faktor u njegovoj karakterizaciji i pravilnoj upotrebi nepostojanje je standardnog protokola za određivanje enzimatske aktivnosti. Općenito je poznavanje enzimatske aktivnosti jedan od ključnih parametara biotransformacija, korištenje prevelike količine enzima može imati negativne ekonomske posljedice za proces, dok, s druge strane, nedovoljna količina može značajno smanjiti produktivnost što posljedično isto negativno utječe na ekonomiku procesa. Cilj ovog rada definiranje je standardnog testa za određivanje volumenske aktivnosti enzima ksilanaze dobivene iz različitih izvora pri čemu su pri razvoju enzimatskog testa u obzir uzeti relevantni procesni parametri kao što su temperatura, pH, koncentracija supstrata, vrijeme inkubacije i koncentracija pufera.

Abstract (english)

Nowadays, the use of enzymes is widespread in many scientific fields such as biotechnology, chemical engineering and process engineering. One of the enzymes that has been widely used in industry in recent decades is endo-1,4-xylanase, which is generally abbreviated as xylanase. A large number of microorganisms produce the enzyme xylanase, among which filamentous fungi are considered the best. However, the search for new sources of xylanases is still intense. Once an enzyme is discovered, named and isolated, one of the major limiting factors in the characterizing the xylanase itself and its proper use is the lack of a standard protocol for determining the enzymatic activities of the xylanase enzyme. In general, the knowledge of enzymatic activity is one of the key parameters of biotransformation. Using too many enzymes can have negative economic consequences for the process, while on the other hand, an insufficient quantity can significantly reduce productivity, which consequently has a negative impact on the economics of the process. The aim of this thesis is to define a standard assay for the determination of the volume activity of xylanase from different sources, while taking into account the relevant process parameters such as temperature, pH, media, substrate concentration, incubation time and buffer concentration in the development of the enzyme assay.
