Abstract

Ovaj rad je sažet pregled značaja i uloge površinski aktivnih tvari u farmaceutskim pripravcima. Površinski aktivne tvari su amfifilne molekule koje posjeduju hidrofilni i hidrofobni dio. Zbog svoje strukture pokazuju posebna fenomenološka svojstva u vodenim otopinama: adsorpcija na granici faza i molekulsko samoudruživanje (micele, vezikule i tekući kristali). Takva svojstva su od velike važnosti u farmaceutskoj industriji, posebno kod istraživanja poboljšanja djelotvornosti lijeka. Za bolju apsorpciju djelatne tvari, a time i za povećanu bioraspoloživost i bolji ukupan farmakoterapijski učinak lijeka, djelatne tvari moraju biti dobro topljive u vodenom mediju te imati dobru propusnost kroz crijevnu membranu s ciljem ulaska u sistemsku cirkulaciju. Primjenjuju se kao nosači lijeka ili sustava za ciljanu isporuku. Postižu željenu koncentraciju lijeka na ciljanom mjestu u tijelu, a također štite tijelo od neželjenih nuspojava. Postoje različite vrste i oblici površinski aktivnih tvari. Ključno je razumijevanje njihovog faznog ponašanja za učinkovitu primjenu u terapijskim sustavima.

Abstract (english)

This thesis is a concise overview of the importance and the role of surfactants in pharmaceutical formulations. Surfactants are amphiphilic molecules that own a hydrophilic and a hydrophobic part. Due to their structure, they show special phenomenological properties in aqueous solutions: phase boundary adsorption and molecular self-aggregation (micelles, vesicles and liquid crystals). This properties are of great interest in pharmaceutical industy, especially at the research of drug efficiency enhancement. For better drug absorption, and therewith for increased bioavailability and better overall pharmacotherapeutic drug performance, drugs have to be well-soluble in an aqueous medium and to own good permeability through the intestinal membrane in order to enter the systemic circulation. They are used as drug carriers or targeted drug delivery systems. They achieve the desired concentration of the drug at the target site in the body, and also protect the body from unwanted side effects. There are different types and forms od surfactants. Understanding the phase behavior is crucial for their effective application in therapeutic systems.
