Abstract

Jedan od osnovnih preduvjeta za život je da organizam efikasno i selektivno katalizira kemijske reakcije koje se u njemu odvijaju. U tome mu pomažu enzimi, makromolekule koje kataliziraju kemijsku reakciju tako da snize energiju aktivacije. Po kemijskom sastavu to su proteini sastavljeni od aminokiselina međusobno povezanih peptidnom vezom. Da nema enzima koji djeluju kao biološki katalizatori, mnoge biokemijske reakcije bile bi prespore ili se uopće ne bi odvijale te su nam upravo zbog toga oni od iznimne važnosti. U usporedbi s drugim katalizatorima, njihovom najvećom prednosti smatra se nenadmašiva selektivnost. Enzimi su prisutni u svim živim organizmima te svaka stanica sintetizira velik broj različitih enzima kako bi provodila svoje metaboličke reakcije. Izbor metode za izolaciju i pročišćavanje enzima ovisi o njihovoj lokaciji, a upravo te metode bit će detaljnije opisane u ovome radu.

Abstract (english)

One of the main prerequisites for life of any organism is that they effectively and selectively catalyze chemical reactions that take place in it. That is achieved by enzymes, macromolecules that catalyze a chemical reaction by lowering the activation energy. By chemical composition, enzymes are proteins composed of amino acids interconnected by a peptide bond. If it were not for enzymes that act as biological catalysts, many biochemical reactions would be too slow or would not take place at all and that is why they are of the utmost importance. Compared to other catalysts, their greatest advantage is their unsurpassed selectivity. Enzymes are present in all living organisms and each cell synthesizes many different enzymes to carry out its metabolic reactions. The choice of method for isolation and purification of a specific enzyme depends on their location, and these methods will be described in more detail in this work.
