Abstract

U ovom radu ispitan je utjecaj osjetljivosti kinetičkih parametara u enzimskoj reakciji između α-ketoglutarne i akrilne kiseline kataliziranoj enzimom
2-sukcinil-5-enolpiruvil-6-hidroksi-3-cikloheksen-1-karboksilne kiseline (MenD) uz tiamin difosfat kao koenzim i magnezijev ion kao kofaktor. Osim produkta, u ovoj reakciji nastaje i nepoznati nusprodukt. Kemijski identitet nusprodukta nije poznat, ali njegovo nastajanje je potrebno uzeti u obzir prilikom razvoja matematičkog modela. Kinetička jednadžba koja opisuje nastanak nusprodukta jednaka je jednadžbi za nastanak produkta. Simulacije napravljene u ovom radu su provedene u Microsoft Excelu korištenjem Eulerove numeričke metode. Dobiveni rezultati simulacija su pokazali da su kinetički parametri koji utječu najviše na konačnu koncentraciju produkta i nusprodukta 𝑉𝑚 i 𝑉𝑚1.

Abstract (english)

In this work, the influence of kinetic parameter sensitivity in the enzymatic reaction between α-ketoglutaric and acrylic acid catalyzed by the enzyme
2-succinyl-5-enolpyruvil-6-hydroxy-3-cyclohexene-1-carboxylic acid (MenD) in the presence of thiamine diphosphate as a coenzyme and magnesium ion as a cofactor was investigated. In this reaction, in addition to the product, an unknown by-product is formed. The chemical identity of the by-product is not known, but its formation had to be taken into account when developing the mathematical model. The kinetic equation describing the formation of the
by-product is equal to the equation for the formation of the product. Simulations presented in this work were carried out in Microsoft Excel using Euler’s numerical method. The obtained simulation results showed that the kinetic parameters with the highest effect on the final concentration of product and by-product are 𝑉𝑚 and 𝑉𝑚1.
