Abstract

Svakodnevna primjena farmaceutika dovela je do njihove konstantne prisutnosti u okolišu, pogotovo u površinskim vodama te tlu i sedimentu. Čak iako su prisutni u niskim koncentracijama mogu značajno poremetiti ekološku ravnotežu, zbog toga je vrlo bitno razviti analitičke metode koje će omogućavati njihovo praćenje u okolišu. Svaka analitička metoda mora osigurati pouzdane rezultate, a upravo to se postiže validacijom metode. U ovom radu validirana je metode tekućinske kromatografije povezana s detektorom s nizom dioda
(HPLC-DAD) za određivanje cefdinira u uzorcima površinskih voda i sintetske otpadne vode. Tijekom validacije određene su izvedbene karakteristike metode (selektivnost/specifičnost, istinitost, preciznost, linearnost, područje primjene, granica kvantifikacije, granice detekcije i stabilnost otopine cefdinira) na temelju čega je zaključeno da je metoda primjenjiva za određivanje cefdinira u vodi. Validacija je provedena s otopinama cefdinira pripremljenim u MilliQ, morskoj, izvorskoj te sintetskoj otpadnoj vodi. Ovo istraživanje i dobiveni rezultati poslužit će kao primjer kako validirati analitičku metodu i dat će uvid u analitičku metodu za određivanje cefdinira koja se može koristiti u budućim istraživanjima.

Abstract (english)

Many pharmaceuticals and personal care products have been detected as aquatic pollutants all over the world. Even if they are present in low concentration, they can have a negative effects on the environment. Therefore, it is very important to develop analytical methods for monitoring pharmaceuticals concentration in the environment. Every analytical method should provide reliable results which is achieved with validation. In this work, high performance liquid chromatography coupled to diode array detector (HPLC-DAD) for the determination of pharmaceutical cefdinir in the environmental waters is validated. During the validation performance, characteristics (selectivity/specificity, accuracy, precision, linearity, working range, limit of quantitation, limit of detection and sample stability) of the method was evaluated and it was concluded that HPLC-DAD method is suitable for its purpose. Cefdinir solutions were prepared in MilliQ, sea water, well water and synthetic wastewater. This research and the results obtained will serve as an example of how to validate the analytical method and provide an insight into a valid analytical method for determining the cefdinir that can be used in future research.
