Abstract

U farmaceutskoj industriji proces kristalizacije danas je jedan od najvažnijih procesa prilikom proizvodnje farmaceutika u čvrstom obliku, odnosno u obliku tableta, što je jedan od najpopularnijih oblika uzimanja lijekova općenito. Stoga, postoji potreba za razvitkom novih tehnika dizajna čestica, kao i potreba za što bržim lansiranjem novih proizvoda na tržište. Sferična kristalizacija nova je tehnika dizajna čestica u kojoj se istovremeno odvijaju procesi aglomeracije i kristalizacije. U ovom radu istraživan je utjecaj dodataka prilikom provedbe kristalizacije iz otopine na granulometrijska svojstva (raspodjelu veličina čestica i oblik) kristala dronedaron hidroklorida. Odabirom pogodnih sustava otapala željelo se utjecati na dobivanje željenih sferičnih aglomerata kristala. Nakon pretraživanja sustava i odabira pogodnih otapala provedena je kristalizacija pri linearnoj brzini hlađenja (12 °C/h) uz dodatak cjepiva. Kao cjepivo korištena je sol dronedaron hidroklorida. Prethodno su određene krivulje topljivosti u određenim otapalima. Na temelju slika dobivenih na svjetlosnom mikroskopu, te SEM-u vidi se da se u sustavima s acetonitirilom dobivaju kristali povoljnijeg obilka koji imaju veću sklonost stvaranju sferičnih aglomerata. Utvrđeno je da se dodatkom različitih otapala utječe na oblik i raspodjelu veličina kristala, ali ne i na njihovu strukturu.

Abstract (english)

In the pharmaceutical industry, the process of crystallization is one of the most important processes in the manufacture of pharmaceuticals in solid tablet form, which is, in general one of the most popular dosage forms. Therefore, there is a need to develop new particle design techniques, and also the need for faster launch of new products on the market. Spherical crystallization is one of such particle design technique in which crystallization and agglomeration process are carried simultaneously. This study investigates the effect of additives in the implementation of crystallization from solution on granulometric properties (particle size distribution and shape) of the obtained dronedarone hydrochloride crystals. Choosing a suitable solvent system is desired to affect obtaining the spherical agglomerates of crystals. After the search and selection of suitable solvent systems crystallization is carried out at a linear speed of cooling (12 °C/h) by adding vaccine. As the vaccine, salt of dronedarone hydrochloride has been used. Solubility of the salt has also been determined previously in certain solvents. Resulting pictures obtained by light microscope and SEM show that systems prepared with acetonitrile have a higher tendency to form agglomerates. It was found that the addition of different solvents affects the shape and the crystal size distribution, but not on crystals structure.
