Abstract

List masline jedan je od nusprodukata maslinarstva koji se akumulira tijekom obrezivanja stabala masline. Masline su prisutne najčešće u Mediteranskim zemljama koje klimatski pogoduju njihovom uzgoju. Lišće masline sadrži znatne koncentracije polifenolnih spojeva koji se izdvajaju i primjenjuju u prehrambenoj, farmaceutskoj te kozmetičkoj industriji. Polifenolnim spojevima pripisuju se mnoge zdravstvene pogodnosti te se upravo zbog toga ekstrakti lišća maslina koriste u liječenju dijabetesa, hipertenzije, bolesti želuca, astme i mnogih drugih bolesti. U kozmetičkoj industriji, ekstrakti lišća maslina koriste se u proizvodima za zaštitu od sunca, anti-age kremama te pripravcima za smanjenje hrapavosti kože i povećanje hidratacije i elastičnosti kože. Najčešće primjenjivana separacijska metoda izdvajanja polifenola iz lišća maslina je ekstrakcija čvrsto-kapljevito. U ovom radu ispitana su različita otapala za ekstrakciju polifenola iz lišća masline. Određene su koncentracije ukupnih polifenola i oleuropelina, polifenolnog spoja koji je prema literaturi najzastupljeniji u listu masline. Koncentracije ukupnih polifenola određene su pomoću UV spektrofotometra, a koncentracija oleuropeina pomoću HPLC uređaja. Ispitana je i mikrobiološka aktivnost ekstrakta kako bi se utvrdilo da li je ekstrakt pogodan za daljnju primjenu u kozmetičkoj industriji.

Abstract (english)

Olive leaf is one of the by-products in olive growing which is accumulated during the pruning of olive trees. Olives are growing in Mediterranean countries due to the suitable climate. Olive leaf contains a significant concentration of polyphenolic compounds which, if separated, can be used in the food industry, pharmacy and cosmetic industry. Polyphenolic compounds are known as health-beneficial and for that reason olive leaf extract is used in diabetes treatment, hypertension, stomach diseases, asthma and many other diseases. In the cosmetic industry olive leaf extract is used in sun protection products and anti-aging creams. What’s more, it improves skin condition by diminishing skin roughness and increasing skin hydration and elasticity. Most commonly used separation method of polyphenolic compounds from olive leaf is solvent-liquid extraction. This study examines different solvents for extraction of polyphenolic compounds. The study found a certain concentration of total phenolic compound and oleuropein, phenolic compound which is most common in olive leaf extract. The concentration of total phenolic compound is determined using UV spectropfotometer, while the concentration of oleuropein is determined using HPLC device. Finally, study examines microbiological activity of the extract with a goal to determine whether the extract was suitable for further use in the cosmetic industry.
