| Abstract | Biorazgradivi materijali su od iznimne je važnosti u inženjerstvu tkiva jer se uklanja potreba za novim kirurškim zahvatom primjenjenog implantata, te osigurava prostor za rast prirodnog tkiva. U procesu razgradnje vrlo je važno postići brzinu koja omogućuje regeneraciju prirodnog tkiva na mjestu oštećenja. Biorazgradivi nosači moraju održavati mehaničku cjelovitost sve dok novo tkivo ne preuzme funkciju i potpuno ih zamijeni. Razgradnju materijala pokreće nekoliko mehanizama, uključujući hidrolizu i enzimsku razgradnju. Najistaknutija skupina biorazgradivih materijala su alifatski poliesteri koji su pokazali primjenu za niz biomedicinskih aplikacija. Poli(ε-kaprolakton) (PCL) je linearni alifatski poliester hidrofobne prirode. PCL je polimer semikristalne strukture kojem se kristalnost smanjuje povećanjem molekulske mase. Njegova dobra topljivost, niska temperatura taljenja (59-64 °C) i dobra kompatibilnost u mješavinama potaknula je opsežna istraživanja njegove potencijalne primjene u biomedicini. Međutim, njegovi nedostatci kao biomaterijala u inženjerstvu tkiva su spora razgradnja i neodgovarajuća mehanička svojstva. Alifatski se poliesteri, time i PCL, razgrađuju jednim ili kombinacijom više mehanizama hidrolizom, enzimskom, mikrobnom i toplinskom razgradnjom. Brzina razgradnje ovisi o nekoliko čimbenika, uključujući stupanj kristalnosti, hidrofilnost, sastav kopolimera, molekulsku masu, molekulnu građu, veličinu i geometriju uzoraka, te uvjete u kojima se razg poput pH i temperature. Razgradnja u okolini je nepromjenjiva in vivo, stoga svojstva polimera u konačnici određuju brzinu razgradnje. Pčelinji vosak (eng. beeswax, BW) je složena smjesa zasićenih i nezasićenih linearnih i složenih monoestera, ugljikovodika, slobodnih masnih kiselina, slobodnih masnih alkohola i manjeg udjela drugih tvari koje proizvode pčele radilice. Izrazito hidrofoban karakter, kristalna struktura i niska temeperatura taljenja (62-65 °C) krakteristike su BW-a. U ovom radu istražen je utjecaj pčelinjeg voska (BW) na brzinu hidrolitičke razgradnje i svojstva poli(ε-kaprolaktona) (PCL) i mješavine poli(ε-kaprolakton)/pčelinji vosak (PCL/BW) uz prisutnost enzima lipaze,koja se nalazi u ljudskom tijelu. Rezultati ukazuju na značajne promjene svojstava i brzine razgradnje poli(ε-kaprolaktona) dodatkom BW, uz enzim lipazu. Na osnovi mjerenja kontaktnog kuta s različitim ispitnim kapljeninama, proračunati su parametri adhezije koji ukazuju na značajnije interakcije između PCL i BW polimera. Hidrofilnost površine PCL/BW mješavina različitog sastava, nakon enzimske razgradnje s lipazom, se povećala što je vidljivo iz smanjenja vrijednosti kontaktnog kuta. U PCL/BW mješavinama BW faza se preferirano smješta na površini. Razgradnja uslijed djelovanja lipaze na PCL polimer i BW odvija mehanizmom površinske erozije. Primjenom površinskih metoda karakterizacije (FTIR-ATR, SEM) utvrđene su strukturne promjene PCL i BW mješavina nakon enzimske razgradnje. Na osnovi dobivenih rezultata DSC mjerenja možemo zaključiti da su temperature taljenja čistih komponenata PCL-a i BW-a vrlo slične, dok se temperature kristalizacije značajno razlikuju. Porast kristalnosti je posljedica apsorbirane vode što se opaža u ranijoj fazi enzimske razgradnje. Apsorbirana voda ima plastificirajuće djelovanje na PCL polimer te čini njegove lance znatno pokretljivijima. BW komponenata PCL/BW mješavine razara kristalnu strukturu PCL polimera. Toplinska razgradnja PCL odvija se u jednom, dok se razgradnja BW odvija u dva stupnja što je utvrđeno primjenom termogravimetrijske analize. Jedan razgradni stupanj PCL/BW mješavina s manjim udjelom voska (do 50 %) ukazuje na njihovu mješljivost. PCL polimer povećava toplinsku stabilnost BW-a. Vrijednosti mehaničkih svojstava čistih komponenata PCL-a i BW-a se značajno razlikuju, a porastom udjela BW faze svojstva PCL/BW mješavina se narušavaju. Tijekom enzimske razgradnje dolazi do sniženja prekidne čvrstoće i prekidnog istezanja, te porasta rasteznog modula, posebice kod BW-a. |
| Abstract (english) | Biodegradable materials are of great importance in tissue engineering because it eliminates the need for a new surgical procedure applied implants, and provides space for the natural tissue growth. In the process of decomposition is very important to achieve the degradation rate that allows the regeneration of the natural tissue at the site of damage. Biodegradable scaffolds must maintain mechanical integrity until new tissue takes their function and completely replaces them. Material degradation runs several mechanisms, including hydrolysis and enzymatic degradation. The most prominent group of biodegradable materials are aliphatic polyesters which have shown useful for a lot of biomedical applications. Poly(ε-caprolactone) (PCL) is a linear aliphatic polyester hydrophobic nature. PCL is semy-crystalline polymer which crystallinity is reduced increasing the molecular weight. It is good solubility, low melting temperature (59-64 °C) and good compatibility in blends initiated numerous researches for its potential applications in biomedicine. However, his limitations as biomaterial in tissue engineering are slow decomposition and inadequate mechanical properties. Aliphatic polyesters, like PCL, degrade by one or a combination of several mechanisms of hydrolysis, enzymatic, microbial and thermal decomposition. The rate of degradation dependent on several factors, including the degree of molecular structure, molecular weightcopolymer composition, crystallinity, hydrophilicity, size and geometry of the sample, and the conditions such as pH and temperature. Environment degradation is unchangeable in vivo, therefore, polymer properties ultimately determine the degradation rate. Beeswax (BW) is a complex mixture of saturated and unsaturated linear and complex monoesters, hydrocarbons, free fatty acids, free fatty alcohol and a lower share of other substances produced by bees. Extremely hydrophobic character, crystal structure and low temperatures range (62-65 °C) are performances of BW. In this work, the influence of beeswax (BW) on the hydrolytic degradation rate and properties of poly(ε-caprolactone) (PCL) and blends of poly(ε-caprolactone)/beeswax (PCL/BW) in the presence of a enzymelipase, which is found in the human body. Results indicate significant changes in properties and degradation rate of poly(ε-caprolactone) whit the addition of BW, in enzyme lipase. For the measurement of contact angle are used different test liquid, calculate the parameters of adhesion that indicate significant interactions between PCL and BW polymer. Hydrophilic surfaces PCL/BW blends of different composition are increased, after enzymatic digestion with lipase, which demonstrated by the decrease in the value of the contact angle. In PCL/BW blends, BW component are preferably placed on the surface. PCL polymer and BW degrade by mechanism of surface erosion. By applying the method of surface characterization (FTIR-ATR, SEM) were determined structural changes PCL and BW mixture after enzymatic degradation. Results based on DSC measurements can be concluded that the melting temperature of pure components PCL and BW very similar, while the temperature of crystallization significantly different. The increase in crystallinity is the result of absorbed water that occurs at earlier stage of enzymatic degradation. Absorbed water has a plasticizing effect on the polymer PCL and makes his chains much more flexible. BW component of PCL/ BW blends destroys the crystal structure of the PCL polymer. Thermal degradation of PCL is in one degree while BW degrade in two degrees, which determined by using thermogravimetric analysis. One degradation degree of the PCL/BW blends with less wax (50 %) indicates their miscibility. PCL polymer increases the thermal stability of BW.The values of the mechanical properties of components PCL and BW are significantly different, and increasing of BW properties PCL/BW blends is damaging. During enzymatic degradation, tensile strength and elongation are reduced, while tensile modules increases, specially for BW. |
