Abstract

U ovom radu prikazana je sinteza, strukturna karakterizacija i in silico analiza derivata 2H-kromen-2-ona (1-4) supstituiranih u položaju 6 bromom, u položaju 3 nitro skupinom ili u položaju 4 arilpropinilnim supstituentima u svrhu ispitivanja njihovog antitumorskog djelovanja. Sonogashira-inom reakcijom 4-klormetil-7-metilkumarina s odgovarajućim arilnim terminalnim alkinima uz Pd-katalizator i CuI kao kokatalizator te uz bazu Et3N sintetizirani su derivati kumarina supstituirani u položaju 4 (3 i 4) arilpropinilnim supstituentima. Strukturna i spektroskopska karakterizacija spojeva provedena je 1H NMR, UV/Vis i fluorimetrijskom spektroskopijom. Farmakološka aktivnost i moguće biološke mete novopripravljenih spojeva predviđene su in silico analizom (PASS).

Abstract (english)

This work describes the synthesis, structural characterization and in silico analysis of 2H-chromen-2-ona derivates (1-4) substituted in position C-6 with bromine, in position C-3 with nitro group or in position C-4 with arylpropynyl substituents in order to determine their antitumor activity. Sonogashira cross-coupling reaction of 4-chloromethyl-7-methylcoumarin with appropriate arylterminal alkynes in the presence of Pd as catalyst, CuI as a cocatalyst and Et3N as a base gave coumarin derivates substituted in position 4 (3 and 4) with arylpropynyl substituents. The structural and spectroscopic characterization of the compounds was carried out by 1H NMR, UV/Vis and fluorimetric spectroscopy. The pharmacological activity and possible biological targets of novel compounds were predicted by in silico analysis (PASS).
