Abstract

Cilj ovog rada bio je optimirati dezintegraciju stanične stijenke L. brevis primjenom metode odzivnih površina (Design expert) u svrhu dobivanja maksimalne aktivnosti alkohol dehidrogenaze u ekstraktu proteina. Dezintegracija se provodila kombinacijom biološke i mehaničke metode tako da se prvo provodila enzimska razgradnja stanične membrane stanice L. brevis primjenom enzima lizozima, a zatim su se stanice izložile ultrazvučnim vibracijama uz prisutnost staklenih kuglica.
Dobiveni enzim u ekstraktu proteina je kinetički karakteriziran u reakciji sinteze kiralnog alkohola (R)-1-feniletanola iz acetofenona. Kinetika reakcije redukcije acetofenona i povratne reakcije oksidacije (R)-1-feniletanola su opisane Michaelis-Menteničinom kinetičkom jednadžbom.

Abstract (english)

The aim of this study was to optimize the disintegration of the cell membrane of L. brevis using response surface metodology to obtain maximal alcohol dehydrogenase activity in the protein extract. The disintegration was carried out by a combination of biological and mechanical methods. First enzymatic degradation of cell membranes cells L. brevis using enzyme lysozyme was carried out, and then the cells was exposed to the ultrasonic vibrations in the presence of glass beads.
The obtained enzyme in the protein extract was kinetically characterized in the reaction of the synthesis of chiral alcohol (R)-1-phenylethanol from acetophenone. The kinetics of the reduction reaction and the reverse reaction of acetophenone oxidation of (R)-1-phenylethanol were described by Michaelis-Menten kinetic equation.
