Abstract

3D-tiskanje omogućuje preciznija pakiranja i doziranja lijekova što je korak više prema budućnosti personaliziranih lijekova. Cilj ovog istraživanja je 3D-tiskanje tableta korištenjem pripremljenih filamenata koji sadrže dronedaron-hidroklorid. Filamenti su pripremljeni pomoću ekstrudera NoztekPro u okviru diplomskog rada studentice Monike Leskovar. Ukupno je pripremljeno jedanaest različitih filamenata pri tri različite temperature (150 ℃, 160 ℃, 170 ℃) i uz korištenje tri različita filamenta poli(vinil-alkohola) (PVA Technology Outlet, PVA Formfutura, PVA Formfutura Atlas). Filamenti su pripremljeni od poli(vinil-alkohola), poli(etilen-glikoa) i dronedaron-hidroklorida. Rađene su dvije formulacije od kojih jedna sadrži 10 %, a druga 20 % dronedaron-hidroklorida. Dobiveni filamenti korišteni su za 3D-tiskanje tableta i epruveta postupkom taložnog srašćivanja. Provedeno je ispitivanje in vitro oslobađanja dronedarona iz dobivenih tableta te test vlačnog ispitivanja za epruvete. Rezultati su pokazali da je kod sva tri poli(vinil-alkohola) koncentracija ukupno oslobođenog dronedarona najveća kod najviše temperature (170 ℃). Profili oslobađanja dronedaron-hidroklorida potkrijepljeni su parametrima odabranih kinetičkih modela. PVA Formfutura i PVA Formfutura Atlas posjeduju veliku mogućnost elastične deformacije i odgovarajuću čvrstoću. Ovim je istraživanjem dokazano da se izborom odgovarajućih procesnih uvjeta i sastava polimernih mješavina mogu pripremiti filamenti kojima će biti tiskane personalizirane tablete.

Abstract (english)

3D printing allows more precise drug packaging and dosing which is a step further towards the future of personalized drugs. The aim of this research is to produce 3D-printed tablets using prepared filaments containing dronedarone hydrochloride. The filaments were prepared using a NoztekPro extruder as part of the graduate thesis of student Monika Leskovar. Eleven different filaments were prepared at three different temperatures (150 ℃, 160 ℃, 170 ℃) and using three different poly(vinyl-alcohol) filaments (PVA Technology Outlet, PVA Formfutura, PVA Formfutura Atlas). The filaments were prepared by using poly(vinyl-alcohol), poly(ethylene-glycol) and dronedarone hydrochloride. Two formulations were prepared containing 10% and 20% of dronedarone. The obtained filaments were used for 3D printing of tablets and test tubes by Fused Deposition Modeling technique. Release profiles of dronedarone from manufactured tablets were obtained using in vitro tests. A tensile test for the tubes was also performed. The results showed that the total concentration of dronedarone released from all three poly(vinyl-alcohol) samples was largest at the highest temperature (170 ℃). Dronedarone hydrochloride release profiles were approximated by parameters of selected kinetic models. PVA Formfutura and PVA Formfutura Atlas are highly elastic with certain material strength. This research shows that by choosing the appropriate process conditions and composition of polymer blends, filaments for printing of personalized tablets can be prepared.
