Abstract

Tehnologija 3D-ispisa u posljednje se vrijeme sve češće javlja pri formulaciji novih dozirnih oblika lijekova. Prepoznate su brojne prednosti poput tiskanja na zahtjev s individualno prilagođenim dozama, modifikacija vremena oslobađanja promjenom geometrije, sposobnost izrade složenih čvrstih oblika, a sve u svrhu izrade lijekova po mjeri pacijenta i kako bi odgovarali upravo njihovim potrebama. Cilj ovog istraživanja je procesom ekstrudiranja taljenjem proizvesti nove filamente s uklopljenom djelatnom tvari dronedaron-hidrokloridom korištenjem komercijalno dostupnih filamenata poli(vinil-alkohola) različitih proizvođača. Uz to, promjenom procesnih uvjeta ekstrudiranja i promjenom udjela djelatne tvari istražiti utjecaj tih parametara na konačan proizvod i njegovu pogodnost za daljnje korištenje u 3D-tiskanju. Od komercijalno dostupnih filamenata korišteni su PVA Atlas i PVA Aqua Solve proizvođača Formfutura te PVA proizvođača Technology Outlet. Mješavine za ekstrudiranje pripremljene su s udjelima DNR-a 10 i 20 % uz dodatak poli(etilen-glikola) kao plastifikatora. Ekstrudiranje taljenjem provedeno je pri temperaturama 150, 160 i 170 °C. Dobivenim je filamentima izmjeren promjer, određen sadržaj te su provedeni testovi bubrenja i vlačnog ispitivanja. Rezultati ukazuju na to da su filamenti PVA Atlas i PVA Aqua Solve s udjelom djelatne tvari 10 % i pri temperaturi ekstrudiranja 150 °C pokazali najbolje karakteristike. Promjeri tih filamenata odstupaju 5,75 % i 4,70 % od ciljanog promjera filamenta. Pokazali su dobre karakteristike bubrenja te nije vidljiv utjecaj erozije. Posjeduju veliku mogućnost elastične deformacije i odgovarajuću čvrstoću. Postotni udio DNR-a ugrađenog u ove filamente iznosi 6,54 i 6,24 %. Ovakav sadržaj DNR-a pripisuje se gubitcima prilikom priprave mješavina te procesa ekstrudiranja. Filament PVA Technology Outlet pokazao je loše karakteristike budući da je ugradnja DNR-a bila neuspješna. Ovim je istraživanjem dokazano kako se izborom komercijalno dostupnih filamenata i udjela djelatne tvari mogu pripremiti filamenti zadovoljavajuće kvalitete za 3D-tiskanje. Također, dokazano je kako temperatura ekstrudiranja utječe na svojstva dobivenih filamenata.

Abstract (english)

Recently, 3D printing technology has become increasingly common in the formulation of novel dosage forms. Numerous advantages are recognized, such as on-demand printing with individually adjusted doses, modification of release time by changing the geometry, the ability to make complex solid forms. All for the purpose of making drugs tailored to the patient and to suit their needs. The aim of this research is to produce new filaments with incorporated active substance dronedarone hydrochloride by hot melt extrusion process using commercially available poly(vinyl alcohol) filaments from different manufacturers. In addition, by changing the extrusion process conditions and changing the content of active substance, investigate the influence of these parameters on the final product and its suitability for further use in 3D printing. Commercially available filaments, PVA Atlas and PVA Aqua Solve from Formfutura and PVA from Technology Outlet were used. Extrusion mixtures were prepared with DNR content of 10 and 20 wt. % with the addition of poly(ethylene glycol) as a plasticizer. Hot melt extrusion was performed at 150, 160 and 170 °C. Filaments were evaluated by measuring the diameter, by determining the drug content and performing swelling and tensile strength tests. The results indicate that filaments PVA Atlas and PVA Aqua Solve with 10 wt.% of DNR and extrusion temperature of 150 °C showed the best characteristics. The diameters of these filaments deviate 5.75% and 4.70% from the target filament diameter. They showed good swelling characteristics and the effect of erosion is not visible. They have high possibility of elastic deformation and adequate strength. The percentage of DNR incorporated in these filaments is 6.54 and 6.24%, respectively. This is attributed to losses during the preparation of mixtures and the extrusion process. The PVA Technology Outlet filament showed poor characteristic as the DNR incorporation was unsuccessful. This research has shown that by selecting commercially available filaments and active substance content filaments of satisfactory quality for 3D printing can be prepared. Also, it has been proven that the extrusion temperature affects the properties of the obtained filaments.
