Abstract

Ovim radom istražena je kompatibilnost djelatne tvari, lurasidon hidroklorida i pomoćne tvari polietilen glikola 4000 Da. Lurasidon hidroklorid ima slabu topivost, lošu bioraspoloživost te usporen početak djelovanja i stoga se ne može dati u hitnim kliničkim situacijama kao što je shizofrenija. Čvrste disperzije pripravljene su metodom isparavanja otapala kako bi se povećala topivost, a samim time i bioraspoloživost djelatne tvari. Ispitivanje kompatibilnosti lurasidon hidroklorida i polietilen glikola provedeno je toplinskim i spektroskopskim analitičkim tehnikama. Diferencijalnom pretražnom kalorimetrijom detektirano je smanjenje temperature taljenja djelatne tvari u čvrstim disperzijama što upućuje na smanjenje veličine kristalita, a samim time i moguću bolju bioraspoloživost djelatne tvari. Dodatno, termogravimetrijskom analizom uočeni su pomaci u maksimalnim temperaturama razgradnje djelatne tvari prema većim vrijednostima što ukazuje na potencijalno postojanje fizikalnih interakcija između djelatne tvari i polimera. Fourier-transformiranom infracrvenom spektroskopijom uočene su promjene u FT-IR spektrima koje potencijalno mogu ukazati na uspostavljanje interakcija između djelatne i pomoćne tvari. Rendgenskom difrakcijskom analizom potvrđeno je postojanje samo kristalne faze djelatne i pomoćne tvari te dobra priprava željenih čvrstih disperzija.

Abstract (english)

With this thesis, drug-excipient (lurasidone hydrochloride-polyethylene glycol 4000), compatibility is studied. Lurasidone hydrochloride owns low solubility, poor bioavailability and slow onset of action and therefore cannot be given in emergency clinical situations like schizophrenia. Solid dispersions were prepared using solvent evaporation method in order to increase solubility and thus drug bioavailability. Drug-excipient compatibility is studied using thermal and spectroscopic analytical techniques. Differential scanning calorimetry detected a decrease in melting temperature of drug in solid dispersions that points to the reduction in crystallite size and hence the possible higher bioavailability of the active substance. Additionally, the shifts in the maximum decomposition temperatures of the drug towards higher values were detected using thermogravimetric analysis indicating the potential physical interactions between drug and polymer. Fourier-transformed infrared spectroscopy revealed changes in FT-IR spectra that might potentially point to the interactions between drug and excipient. Crystal form occurrence for active and excipient substance, and good solid dispersion preparation as well was proven with X-ray powder diffraction analysis.
