Abstract

Kristalizacija je jedna od najvažnijih procesa farmaceutske industrije. Odabir procesnih uvjeta značajno utječe na veličinu, oblik te samu čistoću kristala. Oblik i veličina kristala od presudne su važnosti za formulaciju čvrstih oblika lijeka. Moguće strukturne transformacije kristala izbjegavaju se ispitivanjem i određivanjem termodinamički najstabilnije forme. U ovom radu provedena je kristalizacija uz dodatak antiotapala, a ispitivana djelatna tvar bila je je Omeprazol koji je predstavnik naprednih lijekova za liječenje peptidnih bolesti. Dodatkom različitih antiotapala te za različite brzine dodatka antiotapala uz održavanje konstantne temperature dobiveni su kristali različitih granulometrijskih karakteristika. Za odabrane procesne uvjete ispitan je utjecaj kubnog profila hlađenja na iskorištenje kristalizacije. Dobivenim kristalima određena je raspodjela veličina kristala, pojavni oblik i unutarnja struktura te toplinska karakterizacija.

Abstract (english)

Crystallization is one of the most important processes in the pharmaceutical industry. The choice of process conditions significantly affects the size, shape and purity of the crystal. The shape and size of the crystals are crucial for the formulation of solid drug forms. Possible structural transformations of crystals are avoided by examining and determining the thermodynamically most stable form. In this work, crystallization with the addition of anti-solvents was performed, and the active substance tested was Omeprazole, which is a representative of advanced drugs for the treatment of peptide diseases. Crystals of different granulometric characteristics were obtained by adding different anti-solvents and for different rates of addition of anti-solvents while maintaining a constant temperature. For selected process conditions, the influence of the cubic cooling profile on the crystallization yield was examined. The obtained crystals were used to determine the crystal size distribution, appearance and internal structure, and thermal characterization.
