Abstract

Niska bioraspoloživost novih kemijskih entiteta u farmaceutskoj industriji uzrokovana je njihovom slabom topljivosti u vodi. Dosad, predstavljene su mnoge tehnike za povećanje topljivosti, a time i bioraspoloživosti djelatnih tvari. Obećavajući pristup povećanja topljivosti djelatnih tvari u oralnim dozirnim oblicima je priprava amorfnih čvrstih disperzija. Znanstveni radovi ukazuju na značaj raspodjele veličina čestica u farmaceutskim pripravcima i njezin utjecaj na topljivost i brzinu oslobađanja djelatne tvari. U ovom radu određuju se raspodjele veličina čestica u različitim farmaceutskim pripravcima. Ispituje se njihov utjecaj na topljivost i brzinu oslobađanja djelatne tvari od posebnog interesa.

Abstract (english)

Low bioavailability of new chemical entities in pharmaceutical industry is caused by their poor aqueous solubility. So far, many techniques have been presented to increase the solubility and thus the bioavailability of drugs. A promising approach for drug solubility enhancement in oral dosage forms is preparation of amorphous solid dispersions. Research papers indicate on importance of particle size distribution in pharmaceutical formulations and its effect on drug solubility and release. In this thesis, particle size distributions in various pharmaceutical formulations are determined. Its effect on solubility and release rate of a drug of particular interest is studied.
