Abstract

In vitro ispitivanje otpuštanja djelatne tvari nalazi sve veću primjenu u farmaceutskoj industriji jer je iznimno bitno za osiguranje kvalitete proizvoda, naročito krutih oralnih oblika, tableta ili kapsula. Navedeno ispitivanje pomaže u određivanju optimalnih količina sastojaka potrebnih za postizanje željenih profila otpuštanja lijeka. Djelatna tvar može se oslobađati difuzijskim mehanizmom ili erozijom, te može imati trenutni, produženi ili kontrolirani učinak. U ovom radu ispitano je kako promjena hidrodinamičkih uvjeta utječe na brzinu otpuštanja djelatne tvari dronedaron-hidroklorida iz tablete Multaq. Različiti hidrodinamički uvjeti postignuti su promjenom broja okretaja miješala, pri čemu su temperatura i viskoznost medija te promjer miješala bili konstantni. Promjena koncentracije dronedarona određena je pomoću UV/Vis spektrofotometra mjerenjem apsorbancije. Dobiveni su profili otpuštanja dronedaron-hidroklorida te su opisani kinetičkim modelom. Rezultati su pokazali da se kinetika otpuštanja dronedaron-hidroklorida može opisati Hopfenbergovim modelom koji je prikladan za opis kinetike otpuštanja kod tableta, a podrazumijeva brzinu erozije matrice kao kinetički kontrolirajući mehanizam. Povećanjem Reynoldsove značajke uočen je trend povećanja konstante brzine otpuštanja djelatne tvari.

Abstract (english)

The in vitro release test of the active pharmaceutical ingredient is increasingly applied to the pharmaceutical industry because it is extremely important for the quality assurance of products, particularly solid oral forms, tablets or capsules. This assay helps to determine the optimal amount of ingredients needed to achieve the desired drug release profiles. The active ingredient can be released by diffusion mechanism or erosion and may have a instant, prolonged or controlled effect. This assignment investigates how changes in hydrodynamic conditions affect the release rate of dronedarone hydrochloride from the Multaq tablets. Different hydrodynamic conditions were achieved by varying the impeller rotational speed, whereby the temperature and viscosity of the medium as well as the impeller diameter were kept constant. The change of dronedarone concentration was determined by UV/Vis spectrophotometer by measuring absorbance. Dronedarone hydrochloride dissolution profiles were obtained and described by kinetic model. The results have shown that dissolution kinetics of dronedarone hydrochloride can be described by a Hopfenberg model suitable for the description of release kinetics from tablets, and implies the rate of erosion of the matrix as a kinetic control mechanism. By increasing Reynolds' number, the trend of increasing the release rate of the active substance has been observed.
